How to Use PCR Tubes?
How to Use PCR Tubes in Molecular Biology Experiments?
1. Preparation
Before starting the PCR experiment, it is necessary to disinfect the PCR tube and other experimental equipment. Before disinfection, the PCR tube can be cleaned with 97% hydrochloric acid that has already been disinfected. There are various disinfection methods, usually using 70% alcohol or acetaldehyde solution for disinfection, but be careful not to let the disinfection solution come into contact with the lid of the PCR tube. After thorough disinfection, use sterile items and masks to operate under clean laboratory conditions。2. Add Sample
Add the PCR reaction solution into the PCR tube according to the formula of the PCR reaction system, and this process needs to be very careful to prevent the reaction solution from dripping outside the PCR tube. If the reaction system is properly prepared, excessive sample addition generally does not lead to PCR failure. But it is also necessary to avoid reaction problems caused by excessive addition of template DNA or trace deletions.
3. Cover the PCR tube tightly
After adding the PCR reaction solution to the PCR tube, gently press down on the lid to ensure that the PCR tube lid is completely closed. If
there is a negative control electrophoresis hybridization positive situation, it is necessary to check whether the PCR tube cover is tight enough to confirm special situations such as repeated contamination in the PCR reaction.
4. Label the PCR tube
After adding all experimental samples, the PCR tubes need to be labeled for subsequent processing and analysis. Common labeling methods include numbering, color, different markings, etc。
